乳酸菌素Gassericin T基因的合成及其在大肠杆菌中的融合表达

[摘要] 目的:在大肠杆菌系统中表达有抗菌活性的乳酸菌素Gassericin T。方法:根据乳酸菌素Gassericin T的基因序列,
把Gassericin T的结构基因gatA编码的氨基酸的密码子转换成大肠杆菌偏爱的形式;用人工合成的寡核苷酸片段,通过重
叠PCR法扩增得到gatA片段(倒基因);将合成的 基因插入pGEX一4T一1,构建pGEX一4T一1一gat融合表达载体,转化大
肠杆菌DH5a株,IPTG诱导表达,经超声裂解后获得包涵体蛋白,经溶解、变性、复性处理后获得GST-Gassericin T融合蛋
白;用琼脂扩散法测定其对金黄色葡萄球菌、大肠杆菌、李斯特菌、枯草杆菌等的抗菌活性。结果与结论:采用pGEx一4T一1
融合表达系统在大肠杆菌中表达了有活性的Gassericin T,融合蛋白以包涵体形式存在。复性的融合蛋白对金黄色葡萄球菌
和大肠杆菌有明显的抑制作用,对李斯特菌的抑制作用不明显。

[Abstract]Objective:To express the bacteriocin Gassericin T with antimicrobial activities in E.coli.Methods:DNA frag—
ment encoding the mature peptide of Gassericin T was prepared by three rounds of PCR from a total of six 59 nt oligoes
after modification of the original sequence to the optional codon usage of E.coli and then cloned into pGEX T一1.Clones
carrying pGEX一4T一1一gat was induced by IPTG.The bacteria were bmken down by ultrasonic and the inclusion bodies
obtained underwent denaturation,renaturation and puriifcation to obtain GST-gassericin T fusion protein.The antimicrobial
activity of the fusion protein was assayed by agar diffusion method.Results: Gassericin T was expressed as GST—fusion
protein in pGEX一4T一1 prokaryotic expression system.The fusion protein was present in the lysate as inclusion bodies.
Gassericin T demonstrated antimicrobial activities against gram-positive.s. re and against gram—negative E.coli. Con-
clusion:Recombinant Gassericin T with antimicrobial activities was generated in pGEX一4T一1 fusion expression system.

细菌素是细菌在代谢过程中通过核糖体合成机制产生的一
类具有抑菌活性的多肽或蛋白质组分,产生菌对其细菌素有自
身免疫性【l,21。乳酸菌素是由乳酸菌产生的细菌素,其抑菌范围主
要是与产生菌种属相近的其他细菌【3-5],但也有一些革兰阳性菌
Gassericin T为疏水性肽,只有C端为亲水性,二硫键的形成可
能与其活性有关,但对其作用机理及遗传机制都不是很明确。为
了研究Gassericin T作用的分子机制,我{flAre合成了gatA片
段, 并在大肠杆菌中高效表达了GsT—Gassericin T融合蛋白。
融合表达产物经复性后获得了有抗菌活性的蛋白。
产生的细菌素对革兰阴性菌起抑制作用。乳酸菌已被公认为安
全无毒的细菌。在过去的几年里,人们对乳酸菌细菌素进行了广
泛的基础和应用性研究,主要是因为它们能有效地抑制食品及
1材料和方法
1.1材料
其产品中的病原菌,是潜在的生物防腐剂和饲料添加剂,在食品
和饲料的发酵和储藏过程中起着重要的作用。近年来,随着乳酸
菌素的不断发现,相关的研究也越来越多。乳酸菌素Gassericin
T是一种由加氏乳杆菌(Lactob illus gasseri)SBT2055分泌的广
表达质粒pGEx一4T_1、大肠杆菌DH5a株、金黄色葡萄球菌
ATCC6538株、链球菌、沙门氏菌等标准菌株均由本实验室保存。
谱抗菌的细菌素阎,其相对分子质量为5 400,含有6个ORF。其
中第4个ORF gatA基因(简称gat基因)编码活性蛋白。gatA编
码的蛋白由75个氨基酸残基组成,分子内含2个Cys残基,可
形成分子内的二硫键,在第17和第18位有1个GG基序的切割
限制性内切酶、 DNA聚合酶为TaKaRa公司产品;丙烯酰
胺、N,N2一亚甲双丙烯酰胺、过硫酸胺、SDS、DTT、溴酚蓝、考马斯
