解郁胶囊对抑郁症大鼠行为学、脑神经递质、血液ACTH、

目的:本课题旨在通过实验研究,客观评价解郁胶囊对慢性应激抑郁症大鼠模型脑内单胺类神经递质D
A、5-HT、血中ACTH、CORT及脑内海马神经组织形态学的影响,为临床用药提供客观的理论依据。
方法:取体重200士20g的SD大鼠(SPF级)72只,雌雄各半,普通喂养1周后,以随机数字表法按性别和体重随机分为6组,每组12只,分别为空白组、模型组、解郁胶囊低剂量组、解郁胶囊中剂量组、解郁胶囊高剂量组和蒲郁胶囊组。除空白组给予正常喂养外,其余各组按慢性应激法复制抑郁症模型,时间为21天。实验第22天开始灌胃给药,除空白组、模型组给予生理盐水灌胃外,其余各组动物分别灌胃相应剂量的药物。灌胃连续进行21天,每日1次。实验期间每周称体重1次并观察大鼠行为学。实验第43天,处死所有大鼠,股动脉取血检测ACTH(促肾上腺皮质激素)、CORT(皮质醇);分离右半脑海马组织,匀浆后检测5-HT(5-羟色胺)、DA(多巴胺),左半脑海马组织用4%甲醛固定后作病理组织检查。
结果:①与空白组相比,慢性应激各组大鼠体重及行为学指标均有不同程度改变,表明在本实验条件下,慢性应激可以复制抑郁症大鼠模型。②解郁胶囊各剂量组能够增加实验大鼠的体重及open-field垂直运动实验得分(P<0.05),高剂量能够提高Open-field水平运动实验得分(P<0.05);解郁胶囊高、低剂量能够增加实验大鼠糖水消耗量(P<0.05),高剂量能够减少实验大鼠强迫游泳不动时间(P<0.05)。③解郁胶囊高、中剂量能够不同程度增加实验大鼠脑内5-HT及DA含量(P<0.05)。④解郁胶囊高、中剂量能够不同程度降低实
验大鼠血中ACTH、CORT含量(P<0.05)。⑤解郁胶囊高剂量组能够改变实验大鼠海马CA3区神经元的组织病理学形态,使CA3区神经元结构完整、边缘清晰,细胞排列整齐、紧密,密度增大。 deeply
结论:
1.解郁胶囊具有抗抑郁的作用,这种作用可能是通过调节慢性应激抑郁症模型大鼠脑内单胺类神经递质代谢、下丘脑-垂体-肾上腺轴(HPA轴)功能及修复海马神经元损伤而实现的。
2.解郁胶囊不同剂量对慢性应激抑郁症大鼠的影响存在一定的量效关系,高、中剂量组优于低剂量组,且高剂量组为最优组。

Abstract
Objective:To investigate the effects of JieYu capsule on behavior、concentration of DA,5-HT in Brain Neurotransmitters、concentration of ACTH,CORT in blood 、morphology of hippocampal neuronal organization in brain of chronic stress rat model of deppression underlying it.
Methods:72 SD rats (SPF) weighting 200±20g were divided into six groups bad on gender and weight randomly,including control group,model group,JieYu capsule low do group,JieYu capsule do group,JieYu capsule high dosage group and PuYu capsule group.Each group was compod of 6 male rats and 6 female rats. Besides the control group,the rats of other groups were induced to depression by chronic stressing for 21 days.While the control group and model group were given physiologic saline by intragastric administration at the 22nd day,the other groups were treated with appropriate dos of JieYu capsule and PuYu capsule.Each drug was ud for 21 days,once a day.The ethology of rats was obrved and the weight of rats were measured once a week during this experiment. At the end of the experiment,ACTH and CORT in femoral artery blood were detected. Right brain hippocampal neuronal organization was parated and homogenated for DA (dopamine)、5–HT(5-hydroxytryptamine) detecting. The left brain hippocampal neuronal organization was fixed by 4% formaldehyde for histopathologic examination.
Results:①Compared with blank group,the weight and behavioral index of chronic stress groups were significantly changed.This results indacated that chronic stress can lead to depresd mode in this experimental conditions.②JieYu capsule can increa the weight and experimental score of open-field vertical movement of rats(P < 0.05).The experimental score of the open-field horizontal
movement was significantly incread in JieYu capsule high dosage group (P < 0.05). The sugar water consumption of experimental rats were marked incread in JieYu capsule high and low dos groups (P < 0.05).High dos of JieYu capsule reduce forced swimming fixed time of the experimental rats (P < 0.05).③High and middle dos of JieYu capsule incread the concentration of 5-HT and DA to different extents in brains of the experimental rats (P < 0.05).④High and middle dos of JieYu capsule reduced the concentration of ACTH and CORT to different extents in the blood of the
experimental rats (P < 0.05).⑤The destroy of histopathological form of hippocampal neuronal organization CA3 area of the experimental rat was alleviated in JieYu capsule high dosage group.The results show that the structure of CA3 area neurons was intact and its edge was clear.The arrange of neural cells were more regular and more compact.jal
Conclusion:
1.JieYu capsules can fight against depression through adjusting the metabolism of monoamine neural transmitter and the function of HPA axis in the chronic stress rat model of depression.
