Cells divide many times throughout their lives.But they cannot do it forever.Once they have reached the limits of their reproductive powers,they enter a state called"nescence(衰老)" in which they carry on performing their duties but stop making new copies of themlves.For years it was assumed that,apart from their refusal to divide,nescent cells were otherwi similar in every detail to their replicating compatriots(同胞).
There is mounting evidence,though,that this is untrue.One study in 2016 reported that nescent cells in the kidneys and heart produce a protein that caus nearby healthy tissues to fail
.Another study found that nescent cells contribute to dias like atherosclerosis and arthritis.New work led by Darren Baker, a biologist at the Mayo Clinic in Minnesota,suggests the accumulation of nescent cells within the brains of mice caus the animals to develop neurodegenerative(神经变性的)dias-- and that clearing out the cells can help prevent them.
Working with a team of colleagues,Dr.Baker obtained a population of mice that had been genetically engineered to quickly develop fibrous tangles(纤维团)of protein in their brains.The tangles are associated with the decline in mental abilities caud by dias like Alzheimer's(早老性痴呆病)When the mice were four months old,Dr Baker collected brain tissue from some,and found nescent cells accumulating in the hippocampus, a ahor-shaped region of the brain involved with learning and memory.By merely six months old,they were accumulating in the cerebral cortex(大脑皮层)as well----as were the tangles that are associated with neurological(神经系统的)decline.
To e what role,if any,nescent cells were playing in their decreasing brainpower,Dr.Baker genetically changed some mice such that their nescent cells could be removed with a twice-weekly do of a specific chemical.That left a subgroup of mice that were still likely to have neurolo
gical dias,but which also had their brains rid of nescent cells.By the time the mice reached six months old,the tangles were almost entirely abnt.When the mice were prented with objects they had met before,they approached them without hesitation,as healthy mice should.In contrast,mice who brains were full of nescent cells approached the objects tentatively,as if they had never en them before.Mice are not people.It remains to be en whether clearing exhausted cells from human brains could have similar benefits.Since it is not possible to do genetic engineering on humans beforehand,drugs that can clear out nescent cells will have to be developed instead.But Dr.Baker's results suggest that is worth trying.Indeed,the next project in his lab is to explore whether clearing nescent cells from the brains of mice that are already suffering from the murine(鼠类的)version of Alzheimer's might allow their already damaged brains to recover.
