1. Purpo: To establish a Standard Operating Procedure for and to ensure smooth
progression of the preparation of culture media.
2. Scope of Application: This procedure is applicable for preparation of culture
media.
3. Responsibility: The QC laboratory technicians are responsible for the
implementation of this procedure and the Director of the laboratory
should assume the responsibility of supervision and inspection for
the effective implementation of this procedure.
4. Contents:

4.2 Calculate the required quantities of solid media as needed.

4.4 Dispen the prepared culture media as needed, and put them into the autoclave
for sterilization.
4.4.1 Instruments and appliances: electric furnace, autoclave, balance, triangular flask,
measuring cylinder, test tube.
4.4.2 Procedures for preparation of various media
4.4.2.1 Nutrient Agar Medium: Add 3.2g of the product into 100ml purified water and
mix, and get the solution ready for u by autoclave processing at 121o C for
15 minutes.
4.4.2.2 Ro Bengal Agar Medium: Add 3.05g of the product into 100ml purified water

4.4.2.3 Bile Lacto Medium: Add 3.5g of the product into 100ml purified water and
heat to dissolve; dispen the solution into test tubes and get the solution

4.4.2.5 Eosin Methylene Blue Agar Medium: Add 4.5g of the product into 100ml
purified water and get the solution ready for u by autoclave processing at

4.5 Pour the sterilized media into two plates and incubate the plates at 30 to 35oC for
24 hours; do not u until negative result is confirmed.
5. Storage and management of relevant records: This document is stored in the
