(1)房水抽吸联合激光房角光凝法建立青光眼大鼠模型,重点观察其眼压改变
特征和病理改变特点,探讨其科研应用的可行性。研究结果如下表(图):

上述研究结果发现实验眼激光术后6次眼压均值较术前明显升高,差异有统计学意义(P< 0.01);对照眼眼压较术前无明显变化(表1),实验组可见少数RGCs内灰蓝色Nissl小体减少,甚至消失,而整个细胞染色均匀。光镜下见实验眼较对照眼RGCs数目明显减少
(图4),逆行标记荧光金染色显示结果基本相似(图5)。RGCs死亡从术后7d开始出现,直至术后56d,死亡高峰在术后28~56d(表2)。

上述研究结果发现:青光眼早期伴随着视网膜节细胞死亡并贯穿整个过程。小胶质细胞通过分泌TNF-a和IL-1B等因子诱导炎症反应,加重神经元损伤,促进节细胞的凋亡。特点表现为细胞形态改变,分布改变和比率增加。应用雷公藤甲素等作用于动物模型,均有保护神经节细胞的作用。
其主要原因是抑制小胶质细胞的活性。

雷公藤甲素能够抑制中枢神经系统小胶质细胞活化。因此,本研究将雷公藤甲素用于慢性青光眼大鼠模型,观察RGCs存活情况的改变,探讨雷公藤甲素治疗青光眼的潜在可能性。
