Enzymatic Conversion of Phosphatidylcholine from Milt of Yel-lowtail Seriola quinqueradiata into Phosphatidylrine

Introduction It has been widely recognized that docosahex-aenoic acid(DHA)improved brain function for human.And phosphatidylrine(PS)is also reported as an effective compound for improvement of human brain function.Both of them are com-mercialized as the functional foods in Japan.However,the PS is converted from phosphatidylcholin(PC)of soy bean.The PS that contained DHA in their molecules is not available from edible source.The aim of this study was to prepare DHA-containing PS from cheap edible source.
Materials&Methods:Phospholipids(PL)in yellowtail milt were extracted with ethanol after removal of neutrallipids with acetone. For a conversion of PC into PS,PL solubilid in hexane were incu-bated with acetate buffer containing L-rine,calcium chloride and phospholipa D for24hours.After the incubation,PL composition was determine by HPLC.
Results&Discussion:Major PL in milt of yellowtail was PC.The PL was rich in DHA at sn-2position.The PC of yellowtail milt was more rapidly converted into PS than that of soybean.As for the yields of PS from PC,the PC from yellowtail milt gave lower yield than soybean PC.Under the optimum condition,however,conver-sion ratio of the milt PC into the PS was more than50%.

Keywords:Food starters;proteomics;Enterococcus faecalis The potential u of Enterococcus faecalis in the food industry is a controversial question becau veral strains belonging to this species may be potential dangerous for the human health. The interest of E.faecalis in fermented food industry depend on its lypolitic and proteolytic activities and on its ability to pro-duce bacteriocins active towards bacterial food contaminants.On the contrary other strains of this same species are potentially pathogens being often involved in nosocomial infections.The extra-cellular proteomes of E.faecalis DISAV1022and E.faecalis H1, isolated from a Piedmont chee and from an open surgical wound exudate,respectively,were analyzed.With this approach we would like to analyze differences among strains belonging to the same species but with different habitat origins.
Supernatants of culture of both the strains were recovered in the exponential pha and proteins were precipitated with TCA. Proteomic analys,performed by two-dimensional electrophore-sis coupled with MALDI TOF/TOF mass spectrometry,highlighted the prence of proteins linked to path
皮炎图片和症状ogenicity only in E.faecalis H1:in particular this strain was able to produce an extracel-lular rine protea and the coccolysin which are among the main known virulence factors of this species.Furthermore it express also superoxide dismuta and some glycolytic enzymes probably prent in extracellular broth as moonlighting proteins, potential virulence factors.On the contrary the food strain didn’t produce potentially dangerous extracellular proteins but rather pheromones and other cross-comunication molecules.The expres-sion of rine protea and coccolysin only in E.faecalis H1were confirmed also by phenotypical assays.To understand if pheno-typical differences depend on differential expression of virulence characters or to genetical acquisition of pathogenicity island,PCR analys were performed:extracellular rine protea and super-oxide dismuta genetical determinats revealed to be prent in both the strains while coccolysin one was prent only in E.faecalis H1.

Selenium is a nutritionally esntial element important for men, animals and microorganisms.The human recommended intake is about55g/day.Selenium deficiency may be associated with various chronic dias such as oxidative stress,cardiovascular dias and some kinds of cancer.Since lenium content in plant-bad food depends on its availability from soil,the level of this element in food and feeds varies among regions.There are veral potential options to improve human lenium intake such as soil fertilization and supplementation of food.Development and com-mercialization of organic forms of lenium have initiated a new era in the availability of lenium-enriched products.Selenium-enriched chicken,pork and beef can be produced using organic lenium in the diet of farm animals.The u of Se-enriched yeasts and lactobacilli is also an interesting approach to solve lenium-deficiency:when the microorganisms are added to ice-creams or yogurt,organic form of Se is then relead at gut level.
In this study we focud our attention on a Lactobacillus buchneri strain,already commercialized as probiotic strain,able to over-accumulate lenium during its growth in a MRS modified medium supplemented with sodium lenite(2.19g/l).Our purpo was to understand,by means of a comparative proteomic approach,if le-nium addition could modify protein expression causing variations in the metabolism of the strain.Two-dimensional electrophoresis followed by MALDI-TOF/TOF mass spectrometry was performed on Lactobacillus buchneri26grown in a MRS medium fortified with lenium and in a control condition(same medium without le-nium).The analysis was developed both on the cytosolic and the membrane fraction in the acidic pI range(4-7).From the cytosolic fraction20differentially expresd proteins were detected:15up-regulated and5down-regulated.Image analysis on the membrane gels is actually in progress.
