BALB/c裸鼠与BALB/c小鼠腹腔巨噬细胞吞噬直肠温度

陆军军医大学分数线摘要:目的:比较成年BALB/c裸鼠与BALB/c小鼠腹腔巨噬细胞吞噬功能的差异,为后续裸鼠实验研究提供客观参照。方
法:利用灌洗法提取小鼠腹腔巨噬细胞,并用流式细胞术鉴定F4/80阳性细胞比例。利用流式细胞术和酶标仪检测巨噬细胞吞噬荧光微球判断巨噬细胞吞噬能力。结果:两种小鼠来源腹腔巨噬细胞形态均呈棱形、短杆状或椭圆形等,两组间细胞密度肉眼未见明显差异。流式细胞术鉴定BALB/c小鼠腹腔巨噬细胞F4/80阳性细胞纯度为65.7%。巨噬细胞吞噬荧光微球实验结果显示BALB/c小鼠腹腔巨噬细胞大量吞噬荧光微球,而BALB/c裸鼠来源巨噬细胞吞噬荧光微球显著减少。酶标仪和流式细胞术法比较两种小鼠腹腔巨噬细胞吞噬功能可见BALB/c小鼠腹腔巨噬细胞平均荧光读数显著大于BALB/c裸鼠来源巨噬细胞,差异均有统计学意义(P<0.05)。结论:成年BALB/c裸鼠与BALB/c小鼠腹腔巨噬细胞吞噬功能存在差异,后续裸鼠实验结果分析需考虑巨噬细胞吞噬功能。

Comparison of Phagocytosis of Peritoneal Macrophages in BALB/c Nude Mice and BALB/c Mice Yan Lang, Shi Wen-jing, Tian Yi-jun, Zhang Ji-qian-zhu, Chen Ji-kuai*

Abstract: Objective: To compare the differences in phagocytic function of peritoneal macrophages between adult BALB/c nude mice and BALB/c mice. It provides data for subquent experimental studies in nude mice. Methods: Peritoneal macrophages of mice were extracted by lavage, and the proportion of F4/80 positive cells was identified by flow cytometry. Flow cytometry and microplate reader were ud to detect macrophage phagocytosing fluorescent microspheres to determine macrophage phagocytosis ability. Results: Both mou-derived peritoneal macrophages were prism-shaped, short-rod-shaped, or oval-shaped. There was no obvious difference in the cell density between the two groups. The purity of F4/80 positive cells in peritoneal macrophages of BALB/c mice identified by flow cytometry was 65.7%. The results of macrophage phagocytosing fluorescent microspheres showed that the peritoneal macrophages of BALB/c mice engulfed the fluorescent microspheres in a large amount. BALB/c nude mou-derived macrophages significantly reduced phagocytosis of fluorescent microspheres. Comparing the phagocytosis function of peritoneal macrophages of two mice with microplate reader and flow cytometry, it can be en that the average fluorescence readings of peritoneal macrophages in BALB/c mice are significantly larger than that of BALB/c nude mou-derived macrophages, and the differences are statistically significant. Conclusion: There is a difference in the phagocytic function of peritoneal macrophages between adult BALB/c nude mice and BALB/c mice. Analysis of subquent experimental results in nude mice needs to consider macrophage phagocytosis.
