DISTRIBUTION OF ANTI-GAL、 ANTI-NONGAL、ANTI-NEUGC AND ANTI-NONNEUGC ANTIBODIES IN DIFFERENT HUMAN POPULATIONS

Abstract:
Purpo: To investigate the distribution of anti-Gal, anti-NeuGc, anti-nonGal and anti-nonNeuGc antibodies in different human populations, and to provide experimental evidence for clinical xenograft donors and its application.
Materials and Methods: 84 healthy human rum samples were collected from volunteers from all over the world. The aorta and the whole blood from different transgenic pigs (GTKO/CD46, GTKO/CD46/NeuGcKO) were purchad from Blacksburg Company, USA. Results: (1) Indirect detection of anti-nonGal antibodies and anti-nonNeuGc antibodies in human rum by FCM: ○1The level of anti-nonGal IgM/IgG antibodies in human rum in combination with GTKO/CD46 porcine erythrocytes was higher than that of endothelial cells, but there was no significance (P > 0.05). However, the level of anti-nonoNeuGc IgM/IgG antibody in human rum was significantly lower than that in GTKO/CD46/NeuGcKO porcine erythrocytes, and the difference was significant (P < 0.05). ○2The majority (>95%) of the humans had anti-nonNeuGc IgM/IgG antibody that binding to porcine aortic endothelial cells, but almost 14% to 23% of the human had anti-nonNeuGc IgM/IgG antibody conjugating to porcine erythrocytes ;○3 The level of rum antibody binding to GTKO/CD46 and GTKO/CD46/NeuGcKO transgenic porcine erythrocytes and aortic endothelial cells was low, but the level of human rum antibody with GTKO/CD46/ NeuGcKO transgenic porcine cells was significantly lower than that of GTKO/CD46 (P < 0.05). ○4The anti-non
Gal IgG antibody was significantly higher in the type blood B than that of A, O and AB blood groups (P < 0.05), and the level of anti-nonGal IgG was significantly higher in Eastern Asians than that of other areas (P < 0.05). of Anti-NeuGc IgG was significantly higher than that of IgM (P < 0.001), but had no correlation with age(P > 0.05). There was no significant difference in the expression level of anti-NeuGc IgM antibody between different genders among 56 men and 28 women (P > 0.05), but the level of anti-NeuGc IgG antibody in men was significantly higher than weman (P < 0.05). ○2There was no significant difference in anti-Gal、anti-NeuGc IgM/IgG antibody between different ABO blood groups (P > 0.05).○3The levels of anti-NeuGc IgG in rum of vaccinated with hepatitis B vaccine and BCG vaccine were higher than tho of untreated humans, and the levels of anti-NeuGc IgG in rum of humans vaccinated with typhoid fever were significantly lower than tho of unvaccinated objects (P < 0.05). ○4The levels of anti-NeuGc IgG antibodies in the population from Middle-East were significantly higher than tho in South Asia, Europe and South America (P < 0.05), but anti-NeuGc IgM, anti-nonNeuGc IgM/IgG antibodies had nothing to do with different geographic location (P > 0.05).

Conclusion: The expression of anti-Gal, anti-nonGal, anti-NeuGc and anti-nonNeuGc IgM/IgG antibodies in human rum was different. Its different expression was important to the tissue matching and organ distributing of clinical trial of xenograft transplantation.
