Abstract
Induced pluripotent stem cells (iPSCs) are a unique tool in the field of revolutionary regenerative medicine. It can be the treatment direction of a particular dia. IPS cells can be lf - renewing and can differentiate into many types of cells,thus becoming a potential source of infinite cells in the body. Therefore,the main direction of this rearch is whether iPS cells can provide cell sources for the treatment of oral dias. BMP4 plays an important role in bone formation and development,especially in the bone formation and development of the mouth and limbs. During the development of human embryo, BMP4 is an important signal molecule in the early stage of embryo differentiation. The formation of tooth depends on the expression of BMP4,so as to make tooth germ develop into incisors. In this project,we will explore the methods to solve oral dias by obrving the relationship between the expression of BMP4 and the biological activity of induced pluripotent stem cells in mice.To construct the expression of BMP4 (bone morphogenetic protein- 4,BMP4) lentiviral vector gene and explore its expression of mou induced pluripotent stem cells (iPSCs) effect of biological activity. Methods iPS cell lines stably overexpressing BMP4 were constructed by lentivirus transfection. The activity of cell proliferation was detected by CCK8,and the expression of AMBN,CK14,DSPP,BSP and Runx2 mRNA was detected by Q-PCR. ALP activity was ud to detect the degree of cell differentiation. Results The results of CCK8 showed that the proliferation activity of overexpresd BMP4 could significantly increa the proliferation acti高中军训
vity and ALP activity (P < 0.001) compared with empty vector group. Q-PCR showed that the expression of BMP4 and empty vector group,incread the cell activity of ALP (P < 0.001),Q-PCR showed that the expression of BMP4 in group Runx2 mRNA expression level was significantly incread (P < 0.001). In addition,the relative expression level of AMBNmRNA in the over expression BMP4 group was 4.473 + 0.456,which was 1.435 times higher than that in the empty carrier group (P < 0.001).
