周昭王摘要：PURPOSE: Aminopeptida isolated from Bacillus licheniformis, a preparation process thereof, and a process for producing natural protein using the same enzyme are provided to easily mass-produce natural proteins including human growth hormone. CONSTITUTION: The aminopeptida isolated from Bacillus licheniformis can remove methionine residue in the amino-terminal of peptides or proteins, and has the amino-terminal amino acid quence of lysine - phenylalanine - rine - lysine - lysine -phenylalanine - asparagine - glutamic acid - asparagine - arginine, and the molecular weight of 43 to 47 kDa, wherein the amino acid quence of the amino-terminal of peptides or proteins is methionine-X-proline; and X is phenylalanine. The process for preparing the aminopeptida compris the step of culturing Bacillus licheniformis, centrifuging the cultured medium, concentrating the supernatant, and purifying the aminopeptida from the supernatant, wherein the Bacillus licheniformis is lected from Bacillus licheniformis ATCC 12759, Bacillus licheniformis KCTC 3058, Bacillus licheniformis
KCTC 3045 and Bacillus licheniformis KCTC 1030. The process for producing peptides or proteins having the methionine free amino-terminal compris treating peptides or proteins expresd by a recombinant method with the aminopeptida isolated from Bacillus licheniformis, wherein the recombinant expresd protein is methionyl human growth hormone.
