摘要：Provided are a method for detecting one or more target nucleic acid quences
to be tested by a single tube and a kit thereof. The method compris the steps of: designing specific isothermal amplification primers and fluorescent probe RNHPs for each target nucleic acid quence to be tested, and labeling the fluorescent probe RNHPs for each target nucleic acid quence to be tested with different fluorescences; in the prence of a ribonuclea RNaH, isothermally amplifying each target nucleic acid quence to be tested under the action of a nucleic acid polymera; and forming hybridization chain products labeled with different fluorescences by means of different target nucleic acid quences to be tested, thereby achieving multiplex detection of a plurality of target nucleic acid quences to be tested. In the prent invention, on the basis of an isothermal amplification technique, real-time multiplex isothermal detection can be achieved just by introducing a special probe RNHP and combining same with RNaH2.
