TGF-β1和PDGF-B在成年猴中枢神经系统的分布及脊髓半横断损伤后的表达变化

轻盈的反义词目的:探讨TGF-β1和PDGF-B在成年恒河猴中枢神经系统的分布,及脊髓半横断损伤后不同时相的表达变化。
方法:健康成年恒河猴15只,随机分为5组。其中1组为正常对照组,另外4组在脊髓T11部位制作左侧脊髓半横断损伤模型,动态观察各组动物后肢运动功能变化。模型动物分别于手术后7d、14d、30d、90d行脑和脊髓取材,冰冻切片免疫组织化学SP法染色,显微镜下观察正常对照组TGF-β1和PDGF-B免疫阳性反应物在中枢神经系统的分布概况,计数脊髓前角TGF-β1和PDGF-B阳性神经元数、结果用SPSS11.0统计软件包进行多因素析因方差分析、LSD检验统计学处理。
结果:1.手机投影到墙上 TGF-β1和PDGF-B的阳性反应物在正常对照组恒河猴CNS的分布区域有所不同。在延髓外侧网状核、小脑齿状核可见明显阳性的TGF-β1免疫反应阳性的神经元,下橄榄核可见弱阳性细胞分布;在三叉神经脑桥核、疑核、三叉神经背核及网状结构可见棕黄色的PDGF-B免疫反应阳性的神经细胞;在脊髓TGF-β1和PDGF-B主要分布在灰质前角、中间内、外侧核及胸核神经细胞,阳性反应物亚细胞定位均在细胞浆和突起。此外,脊髓白质也可见胶质细胞呈中等阳性反应。2.猴脊髓半横断术后24h,半横断组左侧后肢肌力为0级,肌张力下降,表现为迟缓性瘫,右侧肌力为3级;伤后14d,脊髓半横断组左后肢肌力均为2级,肌肉萎缩;至伤后90d,左后肢肌力恢复至3~4级。3.脊髓前角TGF-β1和PDGF-B阳性神经元数分别在脊髓半横断损伤后7d、14d下调至最低点, 随着时间延长表达进行性增加,90d时与正常对照组相比较无显著性差异(P>0.05),半横断侧TGF-β衣服变黄1和PDGF-B阳性细胞数比对侧增加(P<0.05)半横断上端阳性神经元数与下端相比无显著性差异(P>0.05),正常对照组与半横断组恒河猴大脑皮质运动区均未见TGF-β1和PDGF-B阳性神经元分布。
结论:1. TGF-β1和PDGF-B在成年猴中枢神经系统均有较广泛的表达,但它们的分布呈现一定的区域性,提示这两种营养因子可能在维持CNS生理功能方面发挥作用,但具体功能
有所不同。

2. 本试验建立了猴脊髓半横断损伤模型,半横断同侧后肢运动功能有一定程度渐进性恢复,提示脊髓半横断伤后有一定运动功能可塑性。

The changes on the expression of TGF-β1 and PDGF-B in central nervous system following spinal cord hemiction in the adult rhesus

Abastract
Objective: To investigate the distribution and changes of TGF-β1 and PDGF-B in the central nervous system of the adult rhesus following spinal cord hemiction.
Method: 15 adult rhesus were divided into five groups randomly. Spinal cord hemiction were performed at T11 level in rhesus, while control group did not receive half-tranction of the spinal cord . Hindlimb function was evaluated and cortical somatonsary evoked potentials(CSEP) were recorded after operation.The aminals received spinal cord hemiction were allowed to survive for 7 days, 14days, 30days and 90days. The caudal and rostral 1 cm spinal cord around the injury site were obtained, and 20 µm thick frozen ctions were cut. By the immunohistochemical SP method, we obrved the distribution of TGF-β1 and PDGF-B immunoreactive cells in the ventral horn of the spinal cord and count tho immunoreactive cells under microscope. The data were statistically analyd with factorial ANOVA and LSD test.
Result: The TGF-β1 and PDGF-B immunopositive neurons were obrved in many areas
in CNS. TGF-β1 immunoreactivity can be en in the cytoplasm and process of medulla oblongata lateral reticular nuclear , cerebellar dentate nucleus and inferior olivary nucleus neuron. PDGF-B immunopositive neurons were obrved in pontine nucleus of trigeminal, ambiguous nucleus , trigeminal dorsal nucleus, reticular formation. TGF-β1 and PDGF-B immunoreactivity mainly distributed in ventral horn neurons , nucleus thoracicus and glia cells in white matter of all normal control and hemiction rhesus. Distinct handicap of locomotor function in hindlimbs was obrved in half-transaction group as soon as SCI. As time going on , locomotor function improved partially. Partial recovery of hindlimb function in adult monkey compared with that of 24 hours postoperatively was en from 14 days to 3month in half-tranction group after SCI. Respectively, the total number of TGF-β1 and PDGF-B immunoreactive neurons in the ipsilateral and contralateral of spinal cord hemiction site 7 days post operation (dpo) and 14dpo on the operative side had declined to the bottom and had significant difference compared with that of normal control group (P<0.05). As the post-operation time gone, the ventral neuron number returned to normal level in 90days post operation.
However, there was no statistic difference between the caudal and rostral spinal cord of all rhesus on the operative side (P>0.05). In addition, TGF-β1 and PDGF-B had the similar expressional trend after the injury. 生命动力按摩椅
Conclusion: A reproducible model of SCI in the nonhuman primate was established. Spontaneous partial recovery of the ipsilateral hindlimb function occurred in monkey of spinal cord hemicted during different periods. It was indicated that there was functional plasticity in the spinal cord after hemiction injury . TGF-β1 and PDGF-B had the similar expressional trend, the immunoreactive positive neurons in ventral horn following spinal cord hemiction reached the most declined level in 7 dpo, 14dpo, and returned to normal level in 90dpo, which showed that TGF-β1 and PDGF-B might involved in the condary injury and function restoretive process after spinal cord injury.
