TB I后脑细胞内Ca2+水平动态变化与CGRP的相关性研究
摘要
目的 探讨钙离子在脑损伤后引起的一系列病理生理改变,进一步探讨钙离子与脑损 伤程度之间的关系及创伤性颅脑损伤(TBI)后钙离子动态变化与降钙素基因相关肽 (CGI心)水平之间的相关性。
方j法 通过对Wistar大鼠硬膜外自由落体打击法来制备颅脑损伤动物模型,采用激光 扫描共聚焦显微镜和酶联免疫吸附实验(ELISA)分别对对照组(36只)和实验组(108 只)测定不同时间点脑细胞内钙离子浓度和血浆中CGRP水平,统计方法采用单因素方差 分析、两因素析因设计方差分析、Pearson简相关分析。
结果对照组大鼠脑细胞内钙离子荧光强度维持在较低的水平,实验组各组大鼠不同 时间点脑细胞内钙离子荧光强度与对照组相比,均有显著性差异(P<0.01),在伤后2小时 左右即有明显升高,约在24小时达到高峰,48小时左右趋于下降。在不同程度脑损伤组 间,重度组脑细胞内钙离子荧光强度水平较轻中度组显著升高口<O.05),随着损伤程度的
加重,大鼠脑细胞内的钙离子荧光强度水平逐渐增高。实验组各组大鼠不同时间点血浆中
降钙素基因相关肽水平与对照组相比,均有显著性差异(P<0.05),实验组内大鼠脑损伤后 血浆中的CGRP水平总体先升高后降低,再缓慢升高。在不同程度脑损伤组间,重度组 CGRP水平较轻中度组显著降低(P<O.05),随着脑损伤程度的加重,大鼠血浆中降钙素基因 相关肽(CGRP)水平逐渐降低。
结论证实脑损伤后脑细胞内存在着钙超载,钙离子在脑损伤后继发性损害中起着重
要的作用;TBI后脑细胞内钙离子水平的动态变化与CGRP之间存在负相关性。
