细胞癌中的表达
摘  要
目的
本课题旨在研究HBV相关肝癌组织中Wnt/β-catenin信号通路中相关因子β-catenin和GSK-3β与HBV相关肝细胞癌发生的关系。
方法
选取20例原发性肝癌患者,均有乙肝病史。按照临床特征如年龄,性别,分化程度,TMN分期,淋巴结转移,HBeAg,HBV DNA 进行统计,经术后病检均为肝细胞癌。通过外科医师在手术室切取这20例肝癌组织和同一患者的癌旁组织。切取后立即将组织标本分类标记后放入-80℃冰箱中冰冻保存,以备提取总RNA及蛋白。通过实时荧光定量聚合酶链式反应(Real-time quantitative polymera chain reaction,QPCR)和Western Blot方法检测肝癌组织和癌旁组织中的β-catenin及GSK-3β mRNA和蛋白的表达水平并进行对比分析。
结果
1. β-catenin mRNA在在肝癌组织相对表达水平高于癌旁组织,差异具有统计学意义(P<0.05);β-catenin 蛋白在肝癌组织的相对表达量高于癌旁组织,差异具有统计学意义水差异有统计学意义(P<0.05)。
2. GSK-3β mRNA在肝癌组织相对表达水平低于癌旁组织,差异

具有统计学意义(P<0.05);失活化型GSK-3β蛋白在肝癌组织的相对表达量高于癌旁组织,差异具有统计学意义(P<0.05)。
结论
守护你一生1.β-catenin及失活化型GSK-3β在肝癌组织中的表达均上调,这为肝癌的分子靶向治疗提供了一定的实验基础。

Abstract
Objective:
The purpo of this study is to study the relationship between Wnt/beta -catenin signaling pathway related factors as beta -catenin and GSK-3 beta and the occurrence of hepatocellular carcinoma (HCC) in HBV-related HCC issue.
Methods:
Twenty cas of patients with primary liver cancer, who were all diagnod with hepatitis B, were enrolled in this study. According to clinical characteristics, such as age, x, differentiation, TMN stage, lymph node metastasis, HBeAg, HBV, DNA,the statistics were taken down. All the patients were diagnod as hepatocellular carcinoma in accordance with  postoperative pathologic examinati
on.Twenty ca of HCC tissues and the paracancerous tissues from the same patient were collected in the operation room.The specimens were labeled and stored in minus eighty Celsius refrigerator for extraction of total RNA and protein. Then, the expression level of mRNA and protein between β -catenin and GSK-3β were detected in the two groups of HCC tissues and

paracancerous tissue,by Quantitative Real-time PCR and Western Blot. The difference of the expression of mRNA and protein in the two groups was analyzed.
Results:
1.The expression level of mRNA of β -catenin in HCC tissues was higher than that in Paracancerous tissue, and the difference was statistically significant (P < 0.05). The expression level of of protein of β -catenin in HCC tissues was higher than that in Paracancerous tissue, and the difference was statistically significant (P < 0.05)
2.The expression level of mRNA of GSK-3β in HCC tissues was lower than that in Paracancerous tissue, and the difference was statistically significant (P < 0.05).The expression level of protein of de
臭狐activated GSK-3β in HCC tissues was higher than that in Paracancerous tissue, and the difference was statistically significant (P < 0.05).
Conclusion:
1.The expression level of β-catenin and deactivated GSK-3βin HCC tissues was up-regulated, which provide certain experimental basis for the molecular targeted therapy of liver cancer.
