荧光增白剂CXT和CBS的物理化学性质及细胞毒性

摘要:目的研究荧光增白剂CXT和CBS的物理化学性质及其对人支气管上皮细胞16HBE和人皮肤角质细胞Hid的生物安全性。方法利用高效液相色谱仪检测紫外吸光度反映荧光增白剂的纯度,利用动态光散射仪检测荧光增白剂分子的尺寸和电荷,使用分光测色仪检测荧光增白剂对棉布的增白效果。以人支气管上皮细胞16HBE和人皮肤角质细胞Hid 为研究对象,利用酶标仪和MTT试剂检测细胞液在490nm处的吸光度反映细胞活力,利用显微镜观察细胞膜表面形态变化,利用流式细胞仪检测细胞液荧光强度反映细胞凋亡情况。通过设立空白对照组,50,100,200m g/mi荧光增白剂CXT和CBS 组,研究荧光增白剂CBS和CXT对这两种细胞的毒理学效应。结果荧光增白剂CXT和CBS主要成分的纯度在97%左右,对棉布均有良好的增白强度,分别达到97.67%和97.67%,其表面具有负电荷和纳米级的分子尺寸。研究结果显示,与空白对照组的细胞相比,荧光增白剂CBS在60m g/mi时16HBE和Hid细胞活力分别降低到80.0%和90.1%,荧光增白剂CXT在100m g/mi时10HBE和Hdd细胞活力分别降低到88.5%和93.2%,但这两种细胞形态均未发生明显变化。细胞凋亡的流式分析结果也证实,即使在200m g/mi的荧光增白剂溶液作
tell me a lie>eqs用下,细胞也未发生明显的凋亡。结论荧光增白剂CXT和CBS表面的负电荷使它们具有良好的水溶性,纳米级的分子尺寸使其染色效果更为持久。在日常使用情况下(洗涤剂中的荧光增白剂的添加量为4-40m/mU,荧光增白剂中无论是CBS还是CXT均不会对呼吸道或者皮肤表面的细胞造成毒性损伤,具有良好的生物安全性。
