2. Applicable Scope: The procedure is applicable to standard operation of passage and isolation
of strain.
3. Responsibilities: QC lab technician shall be responsible for implementation of the procedure,
and lab director shall be responsible for supervising and inspecting the effective
implementation of the procedure.
4.Contents:
4.1 After purchad, the strain shall be prerved in the refrigerator of lab, and managed by the
specially assigned QC lab technicians.
4.2 At the specified time and temperature, inoculate the strain onto the fresh nutrient agar slant
for culturing, conduct passage and inoculation once a month, isolate and purify the strain on the agar plate every three months, and after that, inoculate it again.
4.3 For the cultured strain slant, check whether there is infectious microbe generated, conductmasterbatch
smear microscopy or lect the typical strain and inoculate it on the slant.
4.4 Passage and inoculation
4.4.1 Before passage and inoculation, prepare the fresh nutrient agar slant, and indicate the strain
name, code, inoculation date and passage number.
4.4.2 Take out the strain slant from the refrigerator, keep it at room temperature for 30min, and
then move it into the inoculation room.
4.4.3 Ignite the alcohol lamp, grasp the strain slant and nutrient agar slant with left hand, hold the
inoculating stick with right hand, and burn the metal part of inoculating loop and inoculating stick on the flame through the flame back and forth for three times.
